Structural changes of the rat kidney cortical substance in the long-term period after burn injury of the skin under conditions of HAES-LX-5% infusion

Abstract

The results of an experimental study on structural changes in the rat kidney cortical substance in the long-term period after burn injury of the skin under infusion of HAES-LX-5% solution are presented and analysed in the article. Infusion of HAES-LX-5% causes significant nephroprotective effects, but the kidney cortical substance is characterized by the presence of morphological changes in the components of the glomerular and tubular apparatus. Positive changes include structural signs of the presence of the functional potential of podocytes due to the preservation of cellular pedicels and stabilization of the thickness of the main septum. In rats with skin burns, there are mitoptosis and microautophagic changes in the mitochondria of epithelial cells of the nephron tubules. Microautophagic changes include fusion of lysosomes with small damaged mitochondria or with small isolated fragments of branched mitochondria. Autophagolysosomes formed undergo a step-by-step transformation that can be defined in spatial dimension as a “base apical transformation gradient”. According to this gradient, the transformation of the autophagolysosome occurs in the direction from the electron-dense autophagolysosome in the basal zone of the cytoplasm of the epithelial cells to the vacuole with electron-transparent content in the apical zone of the cytoplasm. Infusion of HAES-LX-5% solution leads to stabilization of the specified gradient, which is a reflection of the protective properties of the solution relative to mitochondria. Mitoptosis plays a leading role in the adaptation of the quantity and quality of mitochondria in epithelial cells of renal tubules to new microenvironmental conditions caused by burn disease. Massive mitoptosis leading to autophagy may result in cell death. The study made it possible to uncover one of the links in the pathogenesis of complications of burn injury of the skin, to find out the peculiarities of morphological changes of the kidney cortical substance and to reveal the positive effect of timely intravenous infusion of HAES-LX-5% on the structure of one of the main organs excreting toxins, which is kidney.
